Ziel der Reise / Objectives of Cruise:

The Pacific Ocean between the subpolar regions of the northern and southern hemisphere stretches over different biogeographic provinces which differ with respect to water masses, hydrography, nutrients and plankton communities. In order to investigate these differences with respect to the composition and functional diversity of the bacterial communities and the dissolved organic matter (DOM) pool a transect from the western edge of the south Pacific subtropical gyre at 30?S to the Bering Sea at 60?N around the 180th longitudinal degree was investigated. Along this transect we visited 19 stations at mean distances of 6 degrees. At 10 of these stations samples were collected by CTD casts between the surface and the sea floor and by a multi corer (MUC) also from surface sediments. Between each deep station only the water column down to a depth of 1000 m was investigated. Further samples were collected by a Bongo net and in situ pump and at stations at the Equator and at 50?N 24 hour time series were analysed as well. The samples from the water column and surface sediment serve to analyse bacterial growth and grazing dynamics and substrate turnover rates by radiotracer techniques, the bacterial community composition (FISH, MAR-FISH) and metagenomics, transcriptomics and metaproteomics by next generation sequencing. In addition, the DOM composition will be assessed by ultrahigh resolution mass spectrometry (FT-ICR-MS). Background parameters such as chlorophyll, particulate organic carbon and nitrogen, DOC and dissolved amino acids and carbohydrates will be analysed as well. A special emphasis was put on the microbial ecology of the prokaryotes in the deep sea below 1000 m depth. Experimental mesocosms set up at stations 2 (south Pacific gyre), 7 (equatorial current) and station 14 (subartic Pacific) served to examine controls of the growth dynamics of the bacterioplankton communities by the availability of DOM, the polysaccharide alginate and by vitamin B12. This large array of microbial, hydrographic and biogeochemical parameters gives us the unique opportunity to comprehensively investigate the functional patterns of prokaryotic life in the biogeographical provinces of the Pacific within one cruise.
